High Throughput Formation of Sub-million Lipid Membrane Arrays with an Asymmetric Lipid Composition
نویسندگان
چکیده
We present a novel method to form more than 10,000 asymmetric lipid-bilayer membranes at a time on micro-chamber arrays. In our method, the arrayed asymmetric lipid-bilayers, where lipid compositions are different between inner and outer leaflets, are formed by injecting several kinds of liquids into a micro-device that has hydrophilic-in-hydrophobic surfaces. The lipid compositional asymmetry is an intrinsic property of bio-membranes, and therefore, this platform extends the versatility of artificial lipidbilayer system, which had been limited to symmetric-bilayer formation, and could contribute to understand the role of lipid compositional asymmetry in cell physiology.

High-throughput formation of lipid bilayer membrane arrays with an asymmetric lipid composition
We present a micro-device in which more than 10,000 asymmetric lipid bilayer membranes are formed at a time on micro-chamber arrays. The arrayed asymmetric lipid bilayers, where lipid compositions are different between the inner and outer leaflets, are formed with high efficiency of over 97% by injecting several types of liquids into a micro-device that has hydrophilic-in-hydrophobic surfaces. ...
متن کاملThe changes in lipid composition of Pythium irregulare LX oomycetes at a stressful situation created with crude oil
Pythium irregulare oomycetes adapts with environmental changes including crude oil concentration by changing the composition of lipids in the cytoplasmic membrane and providing the required characteristics for adaptation in improper and stressful environmental situations. It was found that cultivation of Pythium irregulare LX oomycetes in the nutrient media with different concentrations of crud...
متن کاملPatterning and Composition Arrays of Supported Lipid Bilayers by Microcontact Printing
Fluid-supported lipid bilayers self-assemble on glass and SiO2 surfaces. We have found that it is also possible to assemble fluid bilayers on plasma-oxidized polydimethyl siloxane (PDMS) surfaces. Furthermore, it is possible to transfer or print the supported bilayer from raised PDMS surfaces, such as are typically used for microcontact printing, to fresh glass surfaces creating a supported bil...

The ability to form artificial lipid membranes in a reliable, high-throughput lab-on-a-chip format has the potential to advance membrane protein studies and the development of sensitive molecular biosensors, ultimately impacting on the development of novel and low-cost synthetic approaches to drug screening. Existing methods are limited in terms of their automation, throughput and ease of use. ...
متن کاملSensing-Applications of Surface-Based Single Vesicle Arrays
A single lipid vesicle can be regarded as an autonomous ultra-miniaturised 3D biomimetic "scaffold" (Ø≥13 nm) ideally suited for reconstitution and interrogation of biochemical processes. The enclosing lipid bilayer membrane of a vesicle can be applied for studying binding (protein/lipid or receptor/ligand interactions) or transmembrane events (membrane permeability or ion channel activation) w...
